Hi I am looking for endopeptidase type1 & type 2 and carboxypeptidase H(E) enzymes that are essential in the conversion of proinsulin to insulin. We have a 40 micron seives that we also use. The cells are always kept on ice. The instrument keeps both the sample and the sorted cells at 4 decrees Celcius. We find that lung macrophages just tend to clump more than others. The stirrer of the Facs Aria just swirls the sample around(I think this is a bad design) and does not mix it properly. The Facs Vantage on the other hand has a proper stiirer and mixes the sample better and maybe that is...

hi there! previous members of the lab were expressing a functionally active protein with two his tags: one c-terminal and one n-terminal (pET28 novagen). i thought that one his tag would be better and constructed a vector with only one his tag and... the protein lost its activity!!! do you have any explanation how two 6xhis tags instead of 1 6x his tag can better stabilize a protein??? the protein (around 30kDa) is dissolved in PBS pH7.4...

I had some terrible blockages on the Facs Aria. I was sorting macrophages, from the lung. These macrophages tend to stick together and form clumbs. We do use a 70 micron seive before sorting to take out the clumbs. These cells just aggregate again. The aspiration tubing on the Facs Aria is very narrow as compared to the Facs Vantage.
