Stay far far away from registry cleaners. They do more harm than good.
Your issues have nothing to do with entries in the registry. The registry
is a data table for the OS. Cleaning will have minimal or negative results.

what does the folder contains.. can be that it have viruses which had
linked the file to your computer registry.. thus showing it in use and
can't delete.. run a malwarebytes scans.. antivirus scan can not see the
malwares.. then try to delete the file
